I just need some clarification of general volume usage.

For example, I have a "speaker" icon on my task bar. Then, I have a soundcard volume setting. Then, on top of that all, I have a volume slider on my YouTube video.

What should I use to tweak my volume?

I suppose that really depends on personal preference! I suppose that the easy answer would be to turn the volume all the way up on all but one of the volume 'knobs', and just use that one thereafter. You don't have a volume knob on your speakers?
Your question is terribly funny, this never even occurred to me.
I generally just use whatever is closest to the mouse cursor .
